Shuusuke Amagai arrives in the Soul Society to fill the vacant position of Captain of the 3rd Division, replacing Gin Ichimaru. He initially presents himself as a modest and approachable leader, often acting clumsy or drunk to lower the guards of his subordinates. Despite the initial skepticism from his lieutenant, Izuru Kira, Amagai works hard to unite the fractured squad through teamwork drills.

As the arc progresses, Amagai reveals his true colors as the mastermind behind the conspiracy involving the Kasumiōji Clan. He utilizes a Bakkōtō, a parasitic weapon that negates Shinigami abilities, to fight Captain-Commander Yamamoto. His goal is revenge, believing Yamamoto was responsible for the death of his father years prior. He activates his Bakkōtō's ultimate form, turning his arm into a massive, armored blade to challenge the strongest shinigami.

The conflict ends when the truth is revealed: his father was not murdered by Yamamoto, but had asked to be killed to contain a dangerous Bakkōtō. Devastated by the realization that his vengeance was misguided, Amagai stabs himself with his own blade. He dies apologizing to Kira and his squad, asking for forgiveness for betraying their trust.
